ABSTRACT

The magnetic molecularly imprinted polymers ( MMIPs ) was synthesiZed by suspension polymers technique using ornidaZole ( ONZ) as template, and methacrylic acid ( MAA) and acrylamide ( AA) as the binary functional monomers. The MMIPs were characteriZed by means of scanning electron microscopy ( SEM ) , Fourier transform infrared spectroscopy ( FT-IR ) , vibrating sample magnetometer ( VSM ) , thermogravimetric analysis ( TGA) and ultraviolet-visible ( UV-Vis) spectrophotometer. The results indicated that the adsorption capacity of MMIPs using the binary functional monomers to ONZ was higher than that using single functional monomer or that of its structural analogues. The Scatchard plot revealed that the template polymer system had a two-site binding behavior and the MMIPs exhibited the maximum rebinding to ONZ at 48.96 μmol/g and 10.60 μmol/g.Combined with high performance liquid chromatographic analysis technology, the prepared MMIPs were successfully applied to extract and enrich trace nitroimidaZoles from river water samples with recoveries range of 85.4%-104.3%.The precision of the results was good, and the manipulation of the developed method was simple and fast comparing to other methods.

ABSTRACT

Objective: To investigate the impact of compatibility ratios of rhubarb-gardenia herbal pair and extraction techniques on anthraquinone dissolution, and to explain the bases of different compatibility of rhubarb-gardenia in the prescriptions of Chinese materiamedica from chemical composition. Methods: Anthraquinones were extracted from rhubarb-gardenia herbal pair of different compatibility ratios by water decoction method and ultrasonic extraction with alcohol. Free and total anthraquinones were analyzed by HPLC and the contents of binding anthraquinones were equal to the total minus the free anthraquinones. Results: The contents of various types of anthraquinones extracted by water decoction method increased and binding anthraquinone content increased significantly after rhubarb was compatible with gardenia. This suggested that gardenia was benefit for binding anthraquinone dissolution. Decocting order (Rhubarb cooked first, last, and at the same time) also affected the content of anthraquinones in water decoction method. Separate ultrasonic extraction was benefit for total and free anthraquinone dissolution. Separate ultrasonic extraction was benefit for total and free anthraquinones dissolution. Simultaneous ultrasonic extraction was benefit for binding anthraquinone dissolution. The extraction effect of ultrasonic method was significantly higher than that of water decoction method. Conclusion: Rhubarb-gardenia herbal pair in different compatibility proportion plays different role in different prescriptions because of the different composition and content of anthraquinones, which proves the rationality of the classic formula.

ABSTRACT

OBJECTIVETo discuss the reverse effect of Yinchenhao decoction(YCHD) in dimethyl nitrosamine (DMN)-induced hepatic fibrosis in rats.

METHODThe rat hepatic fibrosis model was established through the intraperitoneal injection with 1% dimethyl nitrosamine (DMN) with a dose of 1.0 mL x kg(-1) x d(-1) for consecutively three weeks, once for the first three days of each. The rats were randomly divided into six groups: the silymarin positive control group (50.0 mg x kg(-1) x d(-1), YCHD high (20.0 g x kg(-1) d(-1)), middle (8.0 g x kg(-1) x d(-1)) and low (3.2 g x kg(-1) x d(-1)) dose groups, the model group and the normal control group. The model group and the normal control group were orally administered with normal saline for consecutively five weeks. The pathologic changes in liver tissues were observed by HE staining. The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), g-glutamyltransferase (g-GGT), hyaluronic acid (HA), laminin (LN), collagen type IV (CIV) and type III procollagen amino terminal peptide (PIIINP) in serum were determined. The metabolite profiling of amino acid and the content of hydroxyproline in liver tissues were also measured.

RESULTCompared with the model group, YCHD high and middle dose groups could significantly reverse the pathologic changes in liver tissues of rats. YCHD could reduce the levels of ALT, AST, gamma-GGT, HA, LN, CIV, PIIINP in serum and the content of hydroxyproline in liver tissues in a dose-dependent manner, and altered the metabolite profiling of amino acid in rat liver tissues.

CONCLUSIONYCHD has the effect in reversing dimethyl nitrosamine induced hepatic fibrosis in rats.
